Dr. Edward A. Botchwey is a Professor in the Wallace H. Coulter Department of Biomedical Engineering at Georgia Tech and Emory University. His research focuses on advancing regenerative medicine and immunoengineering through a convergence approach that integrates various disciplines and techniques for tissue repair and regeneration. Dr. Botchwey’s lab has made significant contributions in investigating S1P signaling in tissue repair and engineering in vivo gradients for microvascular remodeling, as well as developing regenerative immunotherapies using specialized pro-resolving lipid mediators (SPMs) and hydrogel-based delivery systems. This talk will mainly focus on targeting S1P signaling to promote tissue repair and regeneration, including the use of sphingosine kinase inhibitors (SK1i) to reduce local S1P levels and promote muscle fiber regeneration. Additionally, the development and application of ApoM-Fc-S1P, a novel S1P chaperone that enhances S1P receptor 1 signaling, will be discussed, highlighting its role in reducing fibrosis and improving muscle healing, particularly in aged animals. Advanced drug delivery systems such as nanofiber scaffolds and hydrogels, which provide localized and sustained release of therapeutic agents like FTY720, will also be introduced. These delivery systems enhance the efficacy of S1P-targeted therapies by ensuring higher drug concentration at the injury site, facilitating better integration and healing. The presentation will cover the use of ultrasound elastography as a noninvasive imaging technique to monitor tissue stiffness and fibrosis, providing valuable insights into the therapeutic efficacy of these approaches. Dr. Botchwey aims to develop innovative, equity-centered solutions to complex challenges in regenerative medicine, with potential applications in diverse fields such as regenerative immunotherapies, metabolic and hematologic disorders, and cell and tissue transplantation. Additionally, he is actively involved in promoting diversity and outreach through programs like SURE (Summer Undergraduate Research Experience) and ENGAGES (Engaging New Generations at Georgia Tech through Engineering and Science), which aim to provide underrepresented students with research opportunities and inspire the next generation of scientists and engineers. ---- Note: The views of this podcast represent those of my guest(s) and I. A Perspective- Exploring Multi-Factorial Aspects of Quantum Dots- Unveiling Potential Applications?
———————————————————————————————————————————-
Introduction
First, in the realm of quantum dots, the gaze has long been fixed upon their size-dependent optical properties. These nanoscopic entities exhibit intriguing behaviors, where larger uniform nanoparticles offer more space for electron waves, consequently influencing their optical characteristics. Conversely, the confinement of subatomic particles within nanoparticles leads to observable quantized effects, significantly impacting their optical manifestations.
Discussion and Inquiry
Second, delving into the foundational principles of quantum chemistry, we often lean on established concepts like Heisenberg's uncertainty principle. This principle underscores the profound influence of momentum and position on entities at the quantum scale. Size, in essence, is a mathematical expression of position across a dimension or multiple dimensions.
However, what if we were to extend our analysis beyond the size-induced, and quantized-light dependent optical traits of nanoparticles? What if we were to consider their intrinsic and resultant momentum, and how this could affect their optical and temperature properties?
Might such an approach engender a spectrum of radiation with tuned multi-components? Could we better understand and tune the quantized light absorption of colloidal particles through an analysis of their intrinsic and resultant momenta and how that is affect by quantized light? Could we tailor these findings to meet diverse research and clinical needs and demands?
With these questions raised, this beckons us to venture beyond a conventional focus on position or size as a main parameter to adjust in pursuit of understanding quantum dot applications, phenomena, and behavior.
As scientists, it may serve us well to consider the profound implications of how these nanoparticles (quantum dots), potentially adjust or change their behavior within the context of an input of quantized light, when possessing an intrinsic or resultant momentum.

In this brief episode we explore key questions such as:
1. What if we could use genetic evolution and modifying the ubiquitin-proteasome pathway for more targeted therapy in proteinopathies, for example utilizing directed evolution and protein engineering with ubiquitin or regions of the proteasome complex, to include antisense mRNA for key motifs apart of amyloid precursor protein (APP), PSEN1 or SNCA? Could we better target the histopathological hallmarks associated with Alzheimer’s disease, such as the plaques of amyloid-beta, that are indicated in Alzheimer’s disease progression?
2. What if we could use different modified tRNA, or agents in the protein translation pathway, to facilitate therapy for NDD proteinopathies.
In reference to question 1, check out this article on the UPS system in the body: The Ubiquitin–Proteasome System in Immune Cells .

Episode Text:
Cardiolipin (CL), also known as diphosphatidyglycerol, is localized and synthesized exclusively in the mitochondria. This glycerophospholipid was first characterized by Mary Pangborn and McFarlane in 1941. Presently, CL is considered a potential therapeutic target for several neurodegenerative diseases (NDDs). Recent developments in the field of lipidomics indicate that the ratio of monolysocardiolipin-to-native CL is a valuable biomarker for diagnosing NDDs such as Barth Syndrome (BTHS). In this review, the role of lipidomics in understanding the function of CL in NDDs is reviewed.

Organic Chemistry as A Fairy Tale: Once upon a time there as a professor who have twelve princes named: Namio, Structurio and his brother Reactivio, Acidio and his brother Basicio, Nucleophilicio and his brother Electrophilicio, Equilibrio, Stereochemico, Sterico and his brother Electronico and Regioselectivio. Each prince was more complementary than their brother. They all stayed together in the mind-town called Organicochemico. One evening when during the mystical week known as “finale” they all came together to compete at the yearly games. Namio was notoriously a political person who voted for the PLP-s, also known as ( Prinicpal-Locant-Parent Chain- Suffix). Structurio competed in the dance competition showing his routines in their condensed, and bond-line forms. Reactivio participated in the track competition particularly in the race called the Kinetic 500. Acidio and Basicio competed in the meditation competition led by Dr. Bronsted who focuses on obtaining and letting go of your inner proton. Nucleophilicio and electrophilicio helped organized the Kinetic 500, while they both competed in the gymnastic competition with Nucleophilicio stretching the farthest and electophilicio serving as the catcher for the cheerleaders. Equlibrio participated in the annual Le-Chatellier’s fencing competition. Furthermore, stereochemico participated in the art competition focusing on 3D drawings and the arrangement of the objects in space. Sterico and electronico being fierce rivals, competed in the rowing competition with sterico winning at points and electronico winning at points. The boat race is still ongoing. Regioselectivio competed in the map reading competition since he was good at predicting the directions of battles and direction for hikes and journeys. These princes still compete today.. and the story continues.
